To establish an efficient in vitro regeneration protocol for orchardgrass (Dactylis glomerata L.), type of medium, several plant growth regulators and their various concentrations were evaluated in a factorial experiment for callus induction and plant regeneration. Endosperm supported mature embryo were used as explants and optimum response in callus induction was observed on Chu medium with 12 mg/L dicamba ( acid). The highest responding embryogenic callus (15%) was found with Murashige and Skoog medium and 8 mg/L (2,4-dichlorophenoxy acetic acid). |
