Methods The study was designed using an experimental descriptive research method at the laboratory. The study subjects included pediatric patients aged below 16 years old diagnosed with whooping cough and treated at the National Children s Hospital. Results The number of days for positive real-time PCR testing results with whooping cough was 12.4 7.0. The average cycle threshold Ct was 25.4 6.1 cycles. Among 136 samples analyzed for macrolide resistance mutations there were 122 samples obtained results of which 14 samples were identified to have the A2047G mutation on the macrolide-resistant 23S rRNA gene accounting for 11.5 14 122 . Hanoi had the highest number of pertussis cases carrying the A2047G mutation followed by neighboring provinces such as Hung Yen and Bac Ninh. The prevalence of pertussis with the macrolide-resistant A2047G mutation was similar in the group of unvaccinated children and the group of vaccinated children namely 11.6 and 10.0 respectively. Conclusions A total of 17 MLVA genotypes were identified in the study populations of which MLVA genotypes carrying the A2047G mutation included MT104 MT195 MT107 and new type B.
