We studied the liver regeneration from precursor cells induced in mice by a choline-deficient diet supplemented with ethionine (CDE). The diet induced liver inflammation and necrosis, measured by serum alanine aminotransferase activity, ARNm F4/80 concentration. Oval cell accumulation, staining by CK-19, started at day 7 and reached a maximum at day 14. In conclusion, we can induce liver regeneration from oval cells in mice by a CDE diet. * Key words: Liver; Mice; Oval cells; Choline; Ethionine. Introduction The liver is an organ with an almost unlimited capacity to regenerate from mature differentiated cells. However, in severe liver injury or after partial hepatectomy combined with inhibition of mature hepatocyte proliferation, regeneration is achieved through the expansion and differentiation of liver precursor cells, termed oval cells. Oval cells originate from adult stem cells located in Hering duct, form periportal ductular structures that radiate towards the center of the lobules and differentiate into hepatocytes [1]. The mechanisms op responde for the activation of adult stem cells and the expansion and differentiation of oval cells are still largely unknown, but microenvironment and matrix remodeling are of importance [2, 3]. Ductular structures are surrounded by a continuous basement membrane [3] separating oval cells from desminand smooth muscle actin-positive (myofibroblastic) hepatic stellate cells. The basement membrane is thought to play role in oval cell accumulation and migration within the hepatic .
